Kostas and his family do not sound like a typical modern day Greek family. Granted, sons (and daughters) usually remain quite close too their parents, however, when they marry (or become common-law) they usually do embrace (at least out of respect to their partners) their in-laws. Your sister's situation sounds rather disturbing. It's very sad that she abandoned her graduate studies; it's even more disturbing that she wishes to cease all ties with her family. Something is not right with her new life. You owe it to your family to make every attempt to communicate with your sister now while this sad state is still rather current. I assume you have attempted to call and write... however, to no avail; am I right? A trip to Greece is the only way you can actually see your sister, become close with her once again and see where she is coming from. This is a big step, however a very significant one. Try to reason with her. Never allow her to forget how much her family loves her. Best wishes.
